The Mouse VEGF-A Cell Lysate ELISA quantitates Ms VEGF-A in mouse cell or tissue lysates. The assay will exclusively recognize both natural and recombinant Ms VEGF-A. Principle of the method The Mouse VEGF-A solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. VEGF (vaskulärer endothelialer Wachstumsfaktor) ist ein homodimeres, Disulfid-verknüpftes Glykoprotein mit 45 kDa, das an der Angiogenese beteiligt ist und die Progression und Metastasierung von Tumoren fördert. Diese verschiedenen Isoformen binden nachweislich an zwei Tyrosin-Kinase-Rezeptoren flt-1 (VEGFR-1) und flk-1/KDR (VEGFR-2), die fast ausschließlich auf endothelialen Zellen exprimiert werden. VEGF und seine hochaffinitätsbindenden Rezeptoren, die Tyrosinkinasen FLK1 und FLT1, werden für die Entwicklung embryonaler Vaskulaturen als wichtig gehalten. Studien haben gezeigt, dass eine alternativ gespleißte Form von FLT1 ein lösliches Protein produziert, das sFLT1 genannt wird und den vaskulären endothelialen Wachstumsfaktor mit hoher Affinität bindet und eine hemmende Rolle bei der Angiogenese spielt. Erhöhte VEGF-Konzentrationen sind mit dem POEMS-Syndrom (Polyneuropathie, Organomegalie, Endokrinopathie, monoklonale Gammopathie, Hautveränderungen) verbunden, auch bekannt als Crow-Fukase-Syndrom, das mehrere Organe im Körper betrifft.
