Description

The Human ShhN (SHH) ELISA quantitates Hu SHH in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu SHH. Principle of the method The Human SHH solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Sonic Hedgehog (SHH) wird nur während der Embryogenese exprimiert und ist entscheidend für die Ausformung des frühen Embryos. Es wird als das induktive Schlüsselsignal bei der Ausformung des ventralen Neuralrohrs, der anterior-posterioren Extremitätenachse und der ventralen Somititen vermutet. Von drei menschlichen Proteinen, die sequenzielle und funktionelle Ähnlichkeit mit dem Sonic Hedgehog-Protein von Drosophila zeigen, ist dieses Protein diesem am ähnlichsten. Das Protein wird als Vorstufe hergestellt, die autokatalytisch aufgespalten wird; der N-terminale Anteil ist löslich und enthält die Signalaktivität, während der C-terminale Anteil an der Vorläuferverarbeitung beteiligt ist. Noch wichtiger ist, dass das C-terminale Produkt kovalent einen Cholesterinrest an das N-terminale Produkt anbindet, das N-terminale Produkt auf die Zelloberfläche beschränkt und so verhindert, dass es im sich entwickelnden Embryo frei diffundiert. Defekte in diesem Protein oder in seinem Signalweg sind eine Ursache für Holoprosencephalie (HPE), eine Störung, bei der das sich entwickelnde Vorderhirn nicht richtig in rechte und linke Hemisphäre getrennt wird. HPE manifestiert sich durch Gesichtsverformungen. Darüber hinaus wird angenommen, dass Mutationen in diesem Gen oder in seinem Signalweg für das VACTERL-Syndrom verantwortlich sein können, das durch vertebrale Defekte, Analatresie, tracheösophageale Fistel mit ösophagealer Atresie, radiale und renale Dysplasie, Herzanomalien und Extremitätenanomalien gekennzeichnet ist.
