The Mouse Progranulin (GRN) ELISA quantitates Ms Progranulin in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms Progranulin. Principle of the method The Mouse Progranulin solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Prograulin ist das Vorläuferprotein von Granulin, auch als Proepithelin oder PC-Zellwachstumsfaktor bezeichnet. Durch die Spaltung des Signalpeptids entsteht reifes Granulin. Sowohl Granulin als auch das kleinere Spaltprodukt von Granulin reguliert das Zellwachstum. Progranulin stimuliert die Proliferation vieler Epithelzellen, fördert ihr verankerungsunabhängiges Wachstum und verleiht, wenn es überexprimiert wird, Epitheltumorigenizität. Das Progranulin/Epithel-Gen ist multifunktional und übernimmt wichtige konstitutive Rollen in der epitheliale Homöostase und in der reproduktiven, immunologischen und neuronalen Funktion. Progranulin ist konstitutiv in einer Reihe von Epithelien exprimiert, insbesondere in der Haut, im GI-Trakt und im Fortpflanzungssystem. Progranulin wird in Immunzellen in vivo und in bestimmten Neuronen im Gehirn exprimiert.
